Gene-optimized CHO cell line development
to GMP production
Developing a biologic shouldn’t be slowed down by fragmented workflows, complex licensing terms, or uncertainty at critical steps.
Through our collaboration with Demeetra, we offer an integrated, royalty‑free route from cell line development to full GMP manufacturing, built on the genetically optimized CleanCut™ GS CHO platform with transposon/transposase‑based stable integration using a CHO K1 GS‑/‑, suspension‑adapted and animal component‑free cell line. This system comes with a full historical package to facilitate smooth regulatory filing and enables quick generation of high‑performing, stable producer clones and a smooth transition straight into our GMP operations.
This approach reflects what we call Beyond – going further in clarity, preparation, and partnership, so your project moves forward with confidence.
How the process works:
Cell line development
CleanCut™ GS CHO foundation with transposase‑based integration, creating stable producer clones ready for scale‑up.
Upstream and downstream optimization using multi-bioreactor systems and Design of Experiments (DoE) for yield, quality, and reproducibility — engineered to translate smoothly into GMP.

FAQ
Why are CHO cells still the standard host for biologics?
They offer human-compatible glycosylation, decades of regulatory precedent, and the ability to scale from a single-cell clone to thousands of liters. That combination is why the large majority of monoclonal and other antibody formats (bsABs, Fc-fusions) are made in CHO.
Does the CHO background really make a difference?
Yes. Older backgrounds such as DG44 are associated with expression instability and lower titers. GS-engineered CHO K1 lines streamline selection and tend to give higher, more stable productivity. Our partner’s (Demeetra) CleanCut GS line knocks out the GS gene at both relevant loci for more stringent selection, and is offered without downstream royalties or milestone payments.
What are the trade-offs between integration methods?
Random integration is inefficient and demands screening hundreds to thousands of clones, with a real risk of unstable expression. Targeted integration is precise but very low-efficiency and low-copy, which can cap titer. Transposase integration is highly efficient, integrates at high copy in stable sites, and minimizes screening.
Can downstream and analytical work start before the clone is selected?
Yes, we develop the downstream process and analytical methods using pool material in parallel with clone selection, so those workstreams are well advanced by the time the final clone is locked. Clinical-grade manufacturing, however, must be based on the clone-derived master cell bank.
What analytical capabilities do you provide?
As your partner we provide a full panel needed for compliant release testing, in process controls and stability testing for both drug substance and drug product. The panel include methods for identity, purity, potency and safety such as SEC-HPLC, SDS-PAGE, ciEF, ELISA CE and cell based assays. All supporting a program from cell line development through to Phase 3 clinical release.
How is viral safety assured through the process?
Through a layered approach: the cell line is tested virus-free. Our master cell bank reconfirms it; and viral clearance studies confirm that any virus introduced during manufacturing would be removed during purification.
When should a program start planning for GMP and scalability?
As early as the lead candidate stage. Because the cell line and master cell bank are permanent, the choices around host, engineering, raw materials, and process scalability should be made with patient material in mind from the start.
What’s the most common mistake when choosing a manufacturing platform?
the cell line as a near-term decision. You live with your cell line and master cell bank for the life of the product, there is no going back without creating a new product. Choosing a line, raw materials, and a process that can scale and last avoids costly rework later.
